Title: Pycnogenol Enhances Skin Elasticity and Hydration by Boosting Collagen Type I and Hyaluronic Acid Synthase Gene Expression in Women
1. Methodology:
During the study, participants provided demographic data and completed a nutrition questionnaire at enrollment, followed by an 8-day washout phase. Throughout the 12-week supplementation phase, participants received 3 × 25 mg of Pycnogenol daily. Assessments were conducted at three time points: at the start, after 6 weeks (day 49), and after 12 weeks (day 91) of supplementation. Evaluation methods included assessing skin-physiological parameters and analyzing molecular markers from a 4-mm punch biopsy of buttock skin.

2. Skin Hydration
Study on Pycnogenol from pine bark extract showed an 8% increase in skin hydration after 6 weeks of supplementation. However, this improvement wasn’t sustained at the 12-week mark. Short-term benefits were observed, but long-term effects on skin hydration weren’t evident.

3. Skin Elasticity
Skin elasticity was assessed before, at 6 weeks, and at 12 weeks of Pycnogenol supplementation. Consistently improved elasticity throughout the study period confirmed the positive effect of the supplement.

4. Skin Fatigue
R4, a measure of skin fatigue, decreased significantly after both 6 weeks (p < 0.001) and 12 weeks (p = 0.009) of Pycnogenol supplementation. This suggests that Pycnogenol intake may effectively reduce skin fatigue over time.

5. Gene expression of HAS-1, collagen (COL) 1A1 and 1A2.
Mean values ± SE (n = 20) were analyzed with the Wilcoxon signed-rank test. Biopsies taken before and after 12 weeks of Pycnogenol supplementation showed a 44% increase in HAS-1 mRNA expression (p < 0.001), indicating potential skin health benefits. Pycnogenol supplementation significantly increased HAS-1 mRNA expression, suggesting a potential role in skin health improvement.
